After installing this plug-in I ran into the same "Request failed, error RTSP/1.0 500 Internal Server Error" error. The above event approach does fix the iTunes DAAP/Airplay Speak potential conflict but I was not to keen on turning-off Windows Firewall as described earlier.
In my case, on W7, the solution was to leave the Firewall ON then "Allow a program or feature through Windows Firewall" in "Control Panel\All Control Panel Items\Windows Firewall" and add "HSPI_Airplay_Speak.exe" to "Home/Work (Private)"

This feature does not play well with Airplay Speakers because even when iTunes is paused it's still own the connection to the Airplay Speaker for some time, preventing the AirplaySpeak plug-in to send anything. So I would set this feature to No and I would use the :"Event Before" and "Event After" settings from the AirplaySpeak config to disable/enable the speaker before/after the TTS is sent, see screenshots below:

did you try to change the "Require Device Verification" setting like explained in the link?

Could you set the log level to debug, then restart the plugin, and please post here the lines logged when the plugin discovers your AppleTV. Thanks

This is HS3 ver. 3.0.0.68; AirplaySpeak ver. 3.0.0.4.
It worked on my old server (WHS). It was running older beta versions of HS3 and AirplaySpeak (I migrated sometime in December, and am just getting to this issue).
New server is a WHS 2011 VM running on VMware ESXi 5.5. The Windows Firewall is completely disabled in the server VM. No anti-virus loaded during these tests.
I have attached two files that were captured after a clean restart. I then executed an &hs.speak command from the control panel resulting in the errors you see in the files.
